Experiment Objective: How assess the qualification of Plant tissue culture grade Gellan Gum ( CAGGEL MTC ).

Experiment Protocol:


  1. l Amount of precipitate after autoclaving
  2. l Clarity after gelling
  3. l Gel strength after gelling (ability to form a gel)


Reagents/Consumables/Instruments: Beaker, conical flask, balance, magnetic stirrer, Autoclave



Procedure, Observation, and Evaluation:

Prepare solutions of CAGGEL MTC (two samples) and regular food grade gellan gum (one sample) at a concentration of 4 g/L. (Add 2g to 500 mL of water, stir magnetically for 30 minutes, then measure and record the pH). Autoclave the different gellan gum samples directly and observe the three samples post-sterilization for the presence of any precipitate or flocculent matter. (CAGGEL MTC show no precipitate or flocculation.)


Prepare 1/2 MS medium, adding 30g of sucrose per 500 mL of medium. Add 2g each of CAGGEL MTC (two samples) and standard gellan gum (one sample) separately. Stir magnetically for 30 minutes, then adjust the pH to 5.8. Stir magnetically for another 30 minutes, measure the pH again, readjust to 5.8 if necessary. Autoclave all the samples, after the three samples have cooled and gelled, invert the conical flasks to observe gel formation. The two CAGGEL MTC samples will form a gel, whereas the regular food grade gellan gum will fail to form a gel.
